Abstract [en]

Climate change, pollution and human interference have led to a decrease in biodiversity and species being threatened with extinction. Freshwater mussels are more susceptible to environmental changes than most due to them being filter feeders. Freshwater mussels are widespread and important to numerous and diverse ecosystems around the world. Despite this importance, there is a concerning lack of knowledge and, research on the genomes of different freshwater mussels. The aim of this research is to increase the knowledge of the genome for the duck mussel species Anodonta anatina through performing genome assembly. The investigation into the genome of the A. anatina started with sample preparation, continued with DNA extraction, Nanopore sequencing before genome assembly was performed and evaluated. The results of the experiment were the generation of several genome assemblies through the usage of Flye, which were then evaluated by QUAST and compared to other published genome assemblies. The evaluation of the assemblies showed smaller assembly sizes than what was desired based on an estimated genome size of 1.5-2.9 Gb. The estimated genome size range was based on recent published studies on other freshwater mussel species. With the most generous assumption of a genome size at 1.5 Gb, the largest assembly created in this experiment covers less than 33% and therefore did not yield a satisfactory assembly. However, it can potentially serve as a foundation for future research to improve upon with regards to the techniques used and through learning from problems encountered during the experiment.
